چکیده

The intra-subject correlations of three clinically meaningful combinations of serum constituents--(a) potassium, calcium, and albumin; (b) urea, creatinine, and uric acid; and (c) aspartate aminotransferase, lactate dehydrogenase, and alkaline phosphatase--were studied in 11 healthy men. Duplicate serum samples were obtained at 800 h, 1100 h, and 1400 h on five different days. All assays were performed on the AutoChemist Multichannel Analyzer. Correlation coefficients differed significantly among the subjects for the following six pairs of serum constituents: urea and creatinine, urea and uric acid, creatinine and uric acid, aspartate aminotransferase and lactate dehydrogenase, aspartate aminotransferase and alkaline phosphatase, and lactate dehydrogenase and alkaline phosphatase. Nonbiological positive correlation between analytical errors (i.e., errors of two different assays performed on the same specimen) was demonstrated for two of the pairs: potassium and calcium, and aspartate aminotransferase and lactate dehydrogenase. The error correlations of these two pairs of constituents comprised a significant component of the observed intra-subject correlations. Probable reasons for these analytical error correlations are discussed.

Factors contributing to intra-individual variation of serum constituents: 3. Use of randomized duplicate serum specimens to evaluate sources of analytical error.

In study of intra-individual variation of serum constituents, we used analysis of variance to separate biological from analytical variation, the latter being estimated from results for duplicate specimens. However, the replication error term in such a model may grossly overestimate instrumental variability, especially when a significant pre-instrumental error component is present. To separate p...

Factors contributing to intra-individual variation of serum constituents. 1. Within-day variation of serum constituents in healthy subjects.

We evaluated the within-day variation of serum constituents in a group of 11 healthy young men. Twenty-two constituents were assayed at the same time, 19 of them on the “AutoChemist” multi-channel analyzer. The statistical model of analysis-of-variance was used to separate certain factors-subject, time of day, and subject-time interactionfrom the analytical variation. The estimate of analytical...

Individual character of variation in time-series studies of healthy people: II. Differences in values for clinical chemical analytes in serum among demographic groups, by age and sex.

Assessment of the significance of an observed set of serum chemical values for determining a person's state of health requires comparison with a set of defined reference values. We tested the assumption that a reference group of individuals, categorized by age and sex, gives a narrower range of variation than does a larger mixed population. If this were true, the demographic set would be a more...

Factors contributing to intra-individual variation of serum constituents: 5. Short-term day-to-day and within-hour variation of serum constituents in healthy subjects.

We evaluated the variations in some serum constituents in a group of healthy young men for two selected time intervals: short-term day-to-day changes and withinhour changes. In the first case, we used a two-way ANOVA model to compute the main-day effect and the subject-day interaction terms, which were combined to yield the total day-to-day variation. A main-day effect was seen to be statistica...

Biological and analytic components of variation in long-term studies of serum constituents in normal subjects. II. Estimating biological components of variation.

Standard statistical analysis of variance has been used on data from weekly determinations of 15 blood constituents in each of 68 normal subjects, our purpose being to isolate and estimate biological components of observed variation. These components include intra-individual, or “personal” variation, and inter-individual, or “group” variation. The most critical problem, that of separating perso...
